Project title: Validation of 3D osteosarcoma models for anti-cancer drug testing.
Research project. WP1 - Project management and coordination (IMGGE) 1.1. To provide legal, financial and administrative management and coordinate Project activities in order to achieve Project goals and objectives 1.2. To provide logistic support for the organization and implementation of the Project activities within different WPs 1.3. To reinforce a material potential through the acquisition of equipment and consumables 1.4. To ensure delivery of the results and reports as planned WP2 – Establishment of in vivo like model for antitumor drug testing. – first year 2.1. Isolation of patient derived tumor cells 2.2. Generation of 3D model system for osteosarcoma from patient derived tumor cells based on alginate carriers in the form of microbeads 2.3. Treatment with various chemotherapeutic drugs in vivo like conditions (Perfusion bioreactor).
Research project. WP3 - Transcriptomic analysis – library preparation, RNA seq and bioinformatic analysis and validation of targets – second year 3.1. Total RNA isolation 3.2. Library preparation and RNAseq 3.3. Bioinformatic data analysis. Transcriptome analysis will be performed to determine differentially expressed gene sets and their involvement in specific signal pathways. WP4 – Analysis and identification of potential target genes for therapeutic modulation – second year 4.1. High-throughput cell-based assays will be used for screening the responsiveness of osteosarcoma on different bioactive compounds (CERTH)..
Training. Training is one of the major aims of the ICGEB grants programme and full details on the people to be involved and their role, as well as potential benefit to be gained from the training need to be clearly addressed in the proposal. Short-term training visits (up to 3 months) of scientists directly involved in the project for hands-on training to learn or improve a particular technique necessary for the implementation of the project..
Consorcium quality. International collaboration is an essential feature of all ICGEB grants and must be fully integrated into the project. Institute for molecular genetics and genetic engineering (IMGGE) (COORDINATOR) team has a long standing expertise in working with different 2D and 3D cell cultures ’’in vitro’’, RNAseq and transcriptomic analysis . Faculty of technology and metallurgy (TMF) – team has a long standing expertise Elective and advanced courses on special topics in specific bioreactor design and operation, and transport phenomena in biochemical, pharmaceutical, and biomedical engineering. Centre for research and technology HELLAS (CERTH) team has expertise in high-throughput cell-based assays for drug-testing. They are also advanced in generation of cellular models for human diseases using genetically modified human stem cells..

Budget guidelines. The maximum annual contribution from ICGEB is Euro 25,000. Funds can only be used to cover expenditures during the duration of the project (2 yrs) that are directly relevant to the project: Consumable items Scientific literature Laboratory equipment Travel and training.
